MBV9220 – Protein Crystal Spectroscopy
Course content
The course gives and introduction to protein crystal spectroscopy, which is used in combination with protein crystallography to study the structure and function of redox- and metalloproteins. For these types of proteins the structures determined by protein crystallography do not to fully describe the state of these proteins. The essential information that is missing is the oxidation state, spin-state, protonation and in some cases the coordination. This information is provided by the spectroscopic and theoretical methods taught in this course. The course is part of the PhD School in Structural Biology (BioStruct) .
Learning outcome
After completing this course we expect your competence to include the following:
- Know the theory for UV-vis and Raman single-crystal protein spectroscopy, and how to use these methods to study redox- and metalloproteins.
- Have knowledge about the theory for electron paramagnetic resonance (EPR) and theoretical methods (density function theory, QM/MM, quantum refinement) for the study of proteins, and how to use these methods.
- Know how X-rays used to solve the crystal structure of proteins can lead to radiation damage of redox- and metallorproteins, and how to use spectroscopy to monitor these effects.
- Know how to combine these methods for studying protein structure, and how to use them in-situ at the synchrotrons.
- Be able to use the above methods in practice.
Admission to the course
Only students admitted to the course may take part in instruction and tuition and sit for the examination.
Recommended previous knowledge
Basic knowledge of protein structure and protein structure determination.
Teaching
The course comprises for each student of 21 lecture hours and 16 hours of lab-exercises. The course is taught intensively for one week in the beginning of the autumn term every second year.
A selected set of articles should be studied before course start. Estimated time for this activity: one week.
All teaching sessions are obligatory and constitute the curriculum together with hand-outs and pre-defined articles
Examination
Approved participation and laboratory journals. Internet examination through Canvas after course ending.
It will also be counted as one of the three attempts to sit the exam for this course, if you sit the exam for one of the following courses: BIOS9220 – Protein Crystal Spectroscopy
